Reverse Transcription Polymerase Chain Reaction
- Use to detect and quantitate RUNX1::RUNX1T1 fusions arising from t(8;21)
- Use to monitor for MRD in patients with a known history of RUNX1::RUNX1T1 fusions arising from t(8;21) and assess the risk of disease relapse
Testing for RUNX1::RUNX1T1 rearrangements may be useful in the evaluation of acute myeloid leukemia (AML) to aid in classification, inform prognosis, and guide therapeutic decision-making. RUNX1::RUNX1T1 fusion transcript testing may be used to monitor for minimal residual disease (MRD). This test uses reverse transcription polymerase chain reaction (RT-PCR) to detect if a mutant transcript is present.
Genetics
RUNX1::RUNX1T1 fusions arising from t(8;21)
Test Interpretation
Analytic Sensitivity
Limit of quantitation: 1:50,000
Limit of detection: 1/100,000 RUNX1::RUNX1T1 t(8;21)-positive cells
Limitations
- Translocations involving other genes or gene partners will not be identified.
- Levels can only be reliably compared if the same laboratory and same sample type are used.
